1. I just bought a Exo Terra 12"X12"X12" terrarium. Is big enough for two BB Toads?

Yes.

2. Is it better to have two toads or is one O.K.?
(in other words: Will one do better alone or does it need a "play buddy")

They aren't social so the number doesn't matter, provided they have space for each toad.

3. How big of a water dish do I need?

Totally up to you. Just make sure it's not very deep - these toads don't do well in water except when breeding.

4. What's the best substrate to use?

Again, up to you. Coconut fiber (coir) is a good place to start.
